Isothermal amplification, such as hybridization chain reaction (HCR), is a simple and reliable method for detecting signal amplification. However, the hairpin in HCR will not fully participate reaction. And after opened, distance between fluorophore quencher does change much. Therefore, magnification limited. Here, we designed new isothermal amplification named assemble nicked displacement (HANDA), combining strand (SDA) ingeniously. HANDA first triggers through target sequence to form long double-strand DNA (dsDNA) with gaps. Then SDA performed from gap obtain large amount of single-stranded (ssDNA), so achieve purpose double The base had also been optimized. best design rule was found universal applicability. We have demonstrated that combined barcodes can be used trace detection ricin. This provides an effective platform various toxic substances.
